Compound Profile

2,5-Dihydro-2,5-Dimethoxyfuran (CAS 332-77-4) is a dihydrofuran derivative bearing two methoxy groups at the 2- and 5-positions, commercially supplied as a mixture of cis and trans isomers. It is the direct upstream precursor to 2,5-Dimethoxytetrahydrofuran (DMTHF, CAS 696-59-3) — catalytic hydrogenation of DMDF yields DMTHF.

The compound is manufactured by electrolytic methoxylation of furan in methanol solution. A greener photocatalytic route using biomass-derived furan under visible light has also been developed. DMDF serves as a versatile masked 1,4-dicarbonyl equivalent and is a critical industrial intermediate in the tropane alkaloid synthesis chain.

Applications

DMDF is an intermediate in the tropane alkaloid synthesis chain. Key applications include:

  • Atropine Sulfate Synthesis: Upon acid-catalyzed hydrolysis with HCl, DMDF yields 2-hydroxy-1,4-butanedial, which is further hydrogenated and rehydrolyzed to produce succinaldehyde (butanedial) — the key C4 dialdehyde building block in the Robinson tropinone synthesis. Atropine Sulfate is a widely used anticholinergic drug in ophthalmology, anesthesiology, emergency medicine, and organophosphate poisoning treatment.

  • Anisodamine Hydrobromide (654-2) Synthesis: DMDF is also a key intermediate for the production of Anisodamine Hydrobromide, an important anticholinergic agent used to improve microcirculation and treat septic shock, widely prescribed in China and emerging markets.

  • DMTHF Production: Catalytic hydrogenation of DMDF produces 2,5-Dimethoxytetrahydrofuran (DMTHF, CAS 696-59-3), which is itself a major pharmaceutical and pyrrole synthesis intermediate.
